[fimg=]I'd like to know how many total players have played a particular game on BGA, specifically, if you have any ELO ranking at all for a particular game then you count. I don't think this information is readily available so I'm curious what the most accurate estimation would be.

The only way I've found to estimate is to look at a past arena season for a game which tells you how many people participated in said season and their placements. That would give you that number as a minimum + N number of non-arena players which is still vague. Does anybody have any other ways to estimate this figure?

Why? I don’t understand the point. I do know that when I go to manual game creation it tells me how many players online know how to play that game and offers me the chance to invite them

AFAIK no you can't, but the developer can.
From the audience page of the game I developed called Saint Poker, I can see the following:

General statistics
Players who played this game 41 555
Total games played 37136
Game average duration 12mn

The total games played can be found from the game page too. So you can do this for any game.

Palace:
Players who played this game 10 280
Total games played 43636

Detective Poker.
Players who played this game 3 504
Total games played 2211

Not the most popular games, and not much of a correlation between total players and games played

Maybe you can look the number of members in the game players group (In community page, look for a group named: <name of the game> players).
In this group are automatically added all players that played that game. That would still be an estimation because some players may have decided to remove themselves from the group to avoid notifications. Ah and you need to be in the group yourself to see it.

Otherwise maybe you can politely ask the developer of the game if they can provide you the stat.

This I think is the the best estimate without having access to the internal stats. Many people (myself included) don't bother removing themselves from a game's group even if they haven't played it for months and don't intend to.
