It's provably impossible in the first edition simply due to actions required. All board spaces require 2 actions to fill, 1 to pick it up and 1 to place. The castles (dark green tiles) and many buildings give you 1 action back for free but that's still not enough.

In the second edition yellow tile #6 which let's you buy tiles with workers changes this as suddenly boardinghouses giving you 4 workers as well as the yellow tile allowing 4 workers per die suddenly give 2 actions back in some sense for a single one and I've seen games filling the board with those rules (I have myself been 1 tile off but could've managed if not optimizing for points instead).

Interestingly it's actually only a single action missing to be able to fill the base game board.

36 tiles to fill needs 72 actions (take tile and place tile)

* You get 50 normally
* 3 from castles
* 12 from brown buildings
* if you can manage to get all 3 mines down the first round they give you 12 silver = 6 actions

Total 50 + 3 + 12 + 6 = 71

As we begin with 1 silver we're actually extremely close to be theoretically able to fill. Nearly feels intentionally made just impossible.
