Yes, only one side is 4 with circle around it (triggering negative effects when specified on weapon / room / robot).

It’s no surprise they used ALL CAPS to emphasize how important this is, because the game itself fails to convey it. That's actually one of its bad-designed aspects. For instance, there should be 2 points for a full victory (winning and surviving) and 1 point just for surviving (without completing the objective).
0 for a loss.

Another poorly designed aspect: no item with which one could freely set fire to the nest

Regarding Queen death, I'd really like to see the Intruder Help sheet

"Intruder tokens now behave differently."

I've found some low rez pics but nothing great. I get that the queen tokens are like blanks but what else?
